电针对APP/PS1转基因小鼠海马P-糖蛋白和间质液β淀粉样蛋白水平的影响(1)

     摘要 目的:通過观察电针对于APP/PS1双转基因小鼠海马P-糖蛋白表达和脑间质液Aβ水平的影响,探讨电针治疗阿尔茨海默病的作用机制。方法:12只4月龄雄性APP/PS1转基因鼠,随机分为电针组和模型组;6只4月龄雄性C57BL鼠为正常对照组。电针“百会”“涌泉”2周后,采用脑微透析检测技术取小鼠脑间质液,Elisa法检测小鼠间质液Aβ水平;免疫组化法观察小鼠海马P-糖蛋白表达情况。结果:免疫组化结果显示,与正常对照组比较,模型组P-糖蛋白表达明显减少;与模型组比较,电针组的表达相对增加。ELISA检测显示,与正常对照组比较,模型组的间质液Aβ明显减少;与模型组比较,针刺组Aβ水平明显下降。结论:电针可以降低脑间质液Aβ水平,并可以升高海马内P-糖蛋白的表达。

    Effects of Electroacupuncture at Baihui (DU 20) and Yongquan (KI 1) on P-glycoprotein

    Abstract Objective:To observe the effect of electroacupuncture on the level of P-glycoprotein and brain interstitial fluid Aβ in hippocampus in APP/PS1 transgenic mice, and to explore the mechanism of electroacupuncture in the treatment of Alzheimer′s disease. Methods:Twelve male APP/PS1 transgenic mice were divided into electroacupuncture group and model group randomly. Six male C57BL mice were used as normal control group. The level of Aβ in the interstitial fluid of mice was detected by Elisa method after two weeks of electroacupuncture at Baihui (DU 20) and Yongquan (KI 1). The expression of P-glycoprotein in hippocampus was observed by immunohistochemistry. Results:Immunohistochemical results showed that the expression of P-glycoprotein was significantly decreased (P<0.01) in the model group when compared with the normal control group, and the expression of P-glycoprotein in the EA group was significantly increased (P<0.05) compared with the model group. ELISA test showed that the level of brain interstitial fluid Aβ in the model group was lower (P<0.01) than the level in the control group significantly. Compared with the model group, the level of Aβ in the acupuncture group was significantly decreased (P<0.01). Conclusion:Electroacupuncture can decrease the level of brain interstitial fluid Aβ and increase the expression of P-glycoprotein in hippocampus, which may explain why the EA have effects on AD.
